iMethod™ Application for Metanephrines Version 1.1 for Cliquid® Software

The method included is for the rapid analysis of metanephrines in human urine and plasma. Metanephrines can be analyzed in human urine when using the SCIEX LC-MS/MS 3200 or 4000 Series Triple Quad or QTRAP Systems. It also includes a method for the quantitation of metanephrines in human plasma developed on the 4000 and 5500 Series Triple Quad and QTRAP Systems.
Calibration is performed using standard-spiked urine and plasma samples. The methods use three deuterated internal standards to correct for sample and instrument variability. Samples are analyzed in positive mode using the Turbo V Ion Source (electrospray ionization) for maximum sensitivity.

Quantitate low levels of metanephrines
The sample is prepared by simple solid-phase extraction for both urine and plasma matrices, followed by injection onto a Phenomenex Kinetex 2.6 µm HILIC analytical column (50 x 2.1 mm, included) and ionization in the Turbo V™ Ion Source. The analytical run has a dynamic range of 25–1000 ng/mL in urine and 5–4000 pg/mL in plasma, with a total LC run time of eight minutes per sample. Figure 1 shows a representative chromatogram for Calibration Standard 4 (at a concentration of 100 pg/mL for each analyte) in the plasma matrix method.

The methods were evaluated using full calibration curves. The robustness of the methods was tested using multiple replicates of quality control samples. The results showed that the assays were accurate, precise, and reproducible.
