How to use a CTC PAL 3 within Analyst® software?

Answer

  1. Create new hardware profile with CTC PAL 3 in the Analyst® software (1.7.3).
Deactivate all hardware profiles.
Open Analyst Device Driver software 1.4 (ADD).
Click “Configure”.
Choose "CTC Analytics LC" and click “Auto configure”.


Click “Configure” and enter the IP Address 192.168.99.230.

Click "Retrieve Configuration".
Click "OK" and close the "Configure" part.

In Analyst® software, create new hardware profile with MS, ADD and the device type for the required LC module.
Chose “Software Application” and under “Setup Device” the “Analyst Device Driver”.

Note: When you activate the hardware profile, avoid multiple clicks, the activation needs a moment. You will hear the beep, e.g. of Exion Controller first and after 1 or 2 minutes, the sound from CTC PAL 3.
 
  1. Importing Scripts to the ADD.
Before you can use the ADD with CTC PAL 3, you need to import the “Scripts”.
Open the ADD and select “Status.” Choose “Show Script Manager”.

Scripts are saved under Program Files/AB SCIEX/AnalystDeviceDriver/CTCScripts.

Per default, three scripts are available:
  • LC MS Fast Injection,
  • LC MS Standard Injection,
  • LC Injection.
Note: If you are only using the normal “LCMS Tool”, only install the LC MS scripts.

 
  1. Managing Racks.
The racks used by the autosampler need to be defined.
Open the ADD, select “Status”, and choose “Show Rack Position Mapping”.


Note: Choosing in Anaylst® software Batch “Tray Holder” or “Drawer” has no effect. Important is the rack number. In this example, Rack 1 to 3 has 2 different plates. Plate 1 is always next to the CTC X-Y Assembly (part on there the arm is moving). Rack 4 is the “Tray Holder” with 3 plates. Which vial size can be used on which plate, is defined on the "Rack Position Mapping" too.

 
  1. Creating an Acquisition Method.
Start creating an acquisition method by writing the MS and HPLC part first. Save the method under a specific name and close.
Note: “Synchronization Mode” is “Manual/AAO Sync”

Open AAD, click on “Method”. Click on “File” and select "Open from Analyst .dam file".

Select the script you want to use by the pulldown menu.
Specify the missing information in red for the “Setup”, e.g. “Tool”, “Injector”, “Wash Station”, and “Liquid Chromatograph”.

Note: The scripts "LCMS Fast" and “LCMS Standard" are using the “LCMS Tool”, which is the standard tool. The difference between the two scripts is only the numbers of cleaning steps after the injection. The script "LC Injection" will use another tool. Only copy this script if you have the required tool.

Modify the parameters for “Load Sample”, “Inject Sample” and “Post Wash” as required.

For “LCMS Fast Injection”:

Click on “File” and "Save to Analyst .dam file". 

Override the method you have created before in Analyst® software. Close the method in ADD.


Open the acquisition method in Analyst® software. Under Analyst Device Driver information for CTC PAL 3 can be found.


Write your Batch as usual.

Note: Only selecting the “Rack Code” has no effect. Important is the right “Rack Position”.

Open ADD and choose “Status” to see the action the CTC PAL 3 is performing.

Note: This article explains the implementation of CTC PAL 3 into Analyst® software 1.7.3 using Analyst Device Driver Software 1.4. The compatibility with older Analyst® software and Analyst Device Driver Software needs to be verified by reading the related installation guides.
