The next chapter in the SWATH story takes this concept even further by allowing every single MSMS iteration to be acquired and directly correlated to the correct precursor. The data produced by Scanning SWATH® Acquisition therefore contains all the information that would be contained in a multitude of different experimental approaches combined in a single all-encompassing dataset.
SCIEX TripleTOF® platforms are known for their high acquisition speeds and superior data quality even at the fastest scan rates. Scanning SWATH® Acquisition allows the user to exploit these scanning speeds, enabling rapid data-independent acquisition that is compatible with fast chromatography with no method development. Fast scanning quadrupole scan moves an overlapping Q1 window through the m/z range, acquiring all MS/MS fragments, allowing a multidimensional dataset to be collected with more data points.
By scanning through the m/z range instead of stepping, while acquiring MSMS at very fast acquisition rates, Scanning SWATH is able to capture every conceivable MSMS event in a manner where every fragment can be directly correlated to its precursor, meaning that data can be interrogated in a wide variety of ways. The extra dimension of data interpretation achieved through this correlation gives increased confidence in identification for both quantitative and qualitative discovery workflows.
Scanning SWATH produces the ultimate multi-dimensional dataset, containing fully correlated MS and MSMS data on all observable species, which can be processed and interpreted in a variety of ways depending on the goal. The specificity of the approach means that the dataset can be treated quantitatively as a repository of SRMs of every observable precursor, while the ability to correlate these transitions back to their precursor gives improved confidence in identification. Additionally, fast scanning of the quadrupole allows discovery approaches such as precursor ion and neutral loss information to be extracted from the dataset for any observable feature.
