Typically, column temperature is set in the LC method. However, the OptiFlow® ion source has its column compartment directly above the probe on the source itself, so the column temperature field is located in the MS method and can only be adjusted there.

To change the temperature of columns used with the Optiflow ion source, navigate to the MS Method workspace in SCIEX OS software and create a new MS method. The column temperature parameter is in the Source and Gas Parameters section of the MS Method. Below is a screenshot illustrating the location of the column temperature field.

 

The Optiflow column temperature can be set between 5-90°C.

How does one change the default auto-calibrate optiflow column temperature? Which seems to be 25 C, which is highly suboptimal because the well insulated column compartments takes an hour or more to come down to 25 C from, say 80 C. So if using auto-calibrate with optiflow column compartment the acquisition time can get nearly 10 x longer because of the waiting for the column to reach 25 C at every calibration. And then it has to go up to sample method temp again.
