Adjusting Integration Parameters to Exclude Peak Tailing Area in Analyst

Issue Description

Some analytes may display an undesirable amount of peak tailing, and it may not be possible to do further method development after validation to remove the peak tailing. Some of the area associated with the peak tailing may need to be excluded from the total area integrated for more accurate results. 


Resolution

It is not possible to remove the area contributed by peak tailing by defining a cut-off time for integration in Analyst® software; however, it is possible to adjust the amount of noise that is used to define the baseline subtracted from the data points by adjusting the integration parameter, noise percentage.

By adjusting the noise percentage upwards, the peak will be integrated with a higher baseline, generally resulting in smaller peak areas. A noise percentage setting of 40-50% is a good starting value. But the noise percentage can be set to as high as 99%.

To use noise percentage, the IntelliQuan algorithm must be used, and the parameters can be adjusted in the IntelliQuan dialog. This dialog can be opened in the Quantitate mode, by clicking Tools > Settings > Quantitation Integration Algorithm. The IntelliQuan algorithm must be selected. Then select Quant Method Editor Settings.

Once in the IntelliQuan dialog box, select Specify Parameters - MQ III. And then increase the Default Noise Percentage to the desired level. Select OK.
