Adjusting Source/Gas Parameters for Automatic Compound Optimization in Analyst® Software Requires Default Settings Change

Answer

During compound optimization, it is very important to use appropriate source conditions based on the liquid flow rate in the source. For example, the default source parameters (T = 0, GS1 = 20 and GS2 = 0) are usually suitable for low-flow, direct infusions (2-20 µL/min).

In Analyst® software, the automatic compound optimization feature will set the source parameters equal to the default source parameter values, and users do not have the option to change these values directly; however, if the user updates the default values, future compound optimizations will use these new conditions.

Changing the source parameters during manual tuning will not affect the default values used during automatic optimization. For example, if Temp is set to 300 °C during manual optimization, the Temp value would immediately switch to the default of 0 °C upon starting automatic compound optimization. The default values are the same defaults that are present in a new LC/MS acquisition method.
